ABSTRACT

Objective To investigate the biological role of LINC01614 in non-small cell lung cancer A549 cells and its drug resistance-related mechanism. Methods The CRISPR/Cas9 technology was used to construct the A549 cell model with knockdown of LINC01614. Transcriptome sequencing was performed on A549 cells knocked down with LINC01614. We validated the transcriptomic differential genes MCAM and ABCC3 at the gene level and MCAM at the protein level, detected the IC50 changes of A549 cells after knockdown of LINC01614 under the effect of different concentrations of cisplatin, and detected the effect of knockdown of LINC01614 on the migration ability of A549 cells. Results Of the 2 713 DEGs after knockdown of LINC01614, a total of 1 626 genes were up-regulated and 1, 087 genes were down-regulated. GO analysis showed that DEGs were associated with intracellular signaling, cell adhesion, and so on. Meanwhile, the KEGG analysis showed that DEGs were associated with the Wnt signaling pathway, TGF-β signaling pathway, and Rap1 signaling pathway. Selection of drug resistance-associated gene ABCC3 from DEGs for validation with MCAM: qRT-PCR results showed that knockdown of LINC01614 significantly down-regulated the expression of MCAM (P<0.05) and upregulated the expression of ABCC3 on A549 cells (P<0.05). After knockdown of LINC01614, the protein expression of MCAM, was significantly decreased in A549 cells (P<0.05); the IC50 of A549 cells to cisplatin was significantly increased (P<0.05); and the scratch healing rate of A549 cells was also significantly decreased (P<0.05). Conclusion LINC01614 may be associated with the proliferation, invasion, and apoptotic pathways of A549 cells. In addition, LINC01614 may exert its migration ability through MCAM and chemoresistance to cisplatin through ABCC3.

ABSTRACT

Objective To explore the correlation of caries susceptibility of patients with the immediate tooth polish, application of fluoride, and the patients' oral hygiene habits after interproximal enamel reduction (IER) in clinical practice. Methods The present study retrospectively collected 45 cases of IER (18 males and 27 females) in 196 tooth sites. Each sample was examined clinically and radiologically to obtain the incidence of caries, and the outcome was correlated with amount of IER,tooth polish, application of fluoride, and the brushing method and frequency for statistical analysis. Results The present study showed that there was no statistical significance among the amount of IER,the site of IER,the patients' brushing method, and the gender with the susceptibility of caries. However, there was statistical significance among the application of fluoride, tooth polish and brushing frequency with the susceptibility of caries. Conclusions The current research suggests that it is desirable to polish tooth and apply fluoride for patients with IER in clinical practices. In the meanwhile, the clinician should pay attention to patients' oral hygiene habits, especially brushing frequency, due to the positive correlation between the brushing frequency and incidence of caries.

ABSTRACT

The estimation of chronologic age based on the stages of third-molar development was evaluated by using the eight stages (A-H) method of Demirjian and the third-molar development was compared, in terms of sex and age, with results of previous studies. The samples consisted of 291 orthopantomograms from young Chinese subjects of known chronologic age and sex (including 139 males with a mean age of 14.67+/-3.62 y and 152 females with a mean age of 14.85+/-3.70 y). Statistical analysis was performed by employing the Mann-Whitney U-test and the t-test. Regression analysis was conducted to obtain regression formulas for calculating dental age from the chronologic age. Our results showed statistically significant differences (P<0.05) in third-molar development between males and females, at the calcification stages D, E and H. And a strong correlation was found between age and third-molar development in both males (r (2)=0.65) and females (r (2)=0.61). New equations (Age=8.76+1.32 Development stage) for estimating chronologic age were derived. It is concluded that third-molar genesis took place earlier in males than in females. The use of third molars as a developmental marker is appropriate in young adults of Central China. The formula obtained in the present study can be used as a guide for estimation of dental maturity and a standard for age estimation for young adults of Central China.

ABSTRACT

OBJECTIVE:To analyze the main factors related to the adverse drug reactions (ADRs) and the drug-induced diseases induced by Chinese patent medicines commonly used for cardiovascular diseases. METHODS:With ADRs of Chinese medicine as key word, the original papers about Chinese patent medicine-induced ADRs were retrieved from all the literature from 1998 to 2008 in full-text data base of CHKD periodicals for statistical analysis. RESULTS:The common Chinese patent medicines commonly used for cardiovascular diseases might induce ADRs and drug-induced disease. CONCLUSION:Chinese patent medicine-induced ADRs have various categories and manifeste differently clinically, which call for attention from both medical staff and patients.

ABSTRACT

OBJECTIVE:To evaluate the common characteristics and differences of the close herbal prescriptions for coronary heart disease(CHD).METHODS:The close herbal prescriptions for CHD were compared in respect of the compositi-on,pathogenesis,function and indication as well as the clinical application in modern Chinese medicine etc.RESULTS:Great importance should be attached to the differences lie in the 8 groups of close herbal prescriptions for CHD in their clinical application.CONCLUSION:The modern Chinese medicine put forward some new approaches for the treatment of CHD aimed at its patterns of syndrome. In the application of these therapies, syndrome differentiation should be carried out with prudence to choose the most effective therapy in view of the difference of the close prescriptions to achieve a better efficacy.

ABSTRACT

OBJECTIVE: To study the similarity and difference of approximate recipes for edema. METHODS: The approximate clinical commonly used recipes for edema were analyzed in respect of medicine constitute, interpretation of the cause, main functions and clinical application of modern Chinese medicine, etc. RESULTS:The similarity and difference of approximate recipes were found out and their key points in clinical application were confirmed. CONCLUSIONS: It must be careful and considerate to identify the type of edema, afford the reasonable recipes and combine the essence of interpretation of the cause when the new therapy methods are put forward according to modern Chinese medicine. Recipes which are chosen and applied according to their similarity and difference can obtain comparatively sound curative effect.
